If you choose to buy Exclusive Upgrade Tickets through the DLC, you will be able to use them as soon as you are in a main story game.

Is The Exclusive Upgrade Ticket Worth It?
The Exclusive Ticket Upgrade is worth it if you have some extra Spinels on hand as they can change the outcome of a fight depending on what weapon you upgrade.
For example, the Sentinel Nine gets a 5x Critical Rate, making it easy to kill enemies with the handgun or to stagger them for a melee attack.
It all depends on what you upgrade, and Spinels are plentiful if you do Requests in the game but buying the DLC is also a good option for those who want to progress fast.
Since you can only get 2 Exclusive Upgrade Tickets during the main story, it is best that you use them wisely, which means on your main weapons.
It is impossible to get Exclusive Upgrades for every weapon in a single run but you can get a majority of them if you purchase the Exclusive Upgrade Tickets DLC.
If ever you do another playthrough of the game with a New Game Plus, you can get another 2 Exclusive Upgrade Tickets, and so on with every other rerun.
